1,250 mg of NMN a day for up to four weeks caused no severe adverse events in a placebo-controlled trial of 31 healthy adults
Bottom line
The human trials show that nicotinamide mononucleotide (NMN) and nicotinamide riboside (NR) raise blood NAD+ at the doses they tested, and that both were well tolerated for weeks to months. In 8- to 12-week trials, NMN also went with a longer six-minute walk in middle-aged adults (Yi 2023) and a faster short walk and better sleep quality in older adults (Morifuji 2024). A beginner can start from what the trials gave: 250 to 900 mg of NMN a day, or up to 1,000 mg of NR, taken daily for about 8 to 12 weeks. Results on function were mixed across the wider set of trials.
How people took it in the trials
Pick the form you will take every day. The trials gave NMN by mouth: 300 to 900 mg in one daily dose (Yi 2023), one 125 mg capsule twice a day for a 250 mg daily total (Katayoshi 2023), 1,250 mg once a day for up to four weeks (Fukamizu 2022), and two 500 mg tablets twice a day for 28 days (Pencina 2023). No human trial in our evidence compared delivery forms, so the choice between liposomal, powder, sublingual or enteric forms comes down to convenience and value.
Start at the dose the trials used. 250 to 900 mg of NMN a day is the range most of the trials used. At 250 mg a day, older adults had a shorter 4-metre walking time than placebo after 12 weeks (Morifuji 2024), and in a trial of 36 healthy middle-aged adults, the NMN group had higher serum levels of a NAD+ metabolite than placebo (Katayoshi 2023). In a trial of 80 healthy middle-aged adults, 300, 600 and 900 mg a day each raised blood NAD+, with the highest levels and longest walking distances at 600 and 900 mg (Yi 2023). A 2026 meta-analysis covered trials from 250 to 2,000 mg a day (Yang 2026). For NR, the trials reached 1,000 mg a day (Conze 2019).
Take it at a steady time each day. Most of the trials gave one dose a day. The one trial that tested timing, in 108 older Japanese adults given 250 mg of NMN or placebo, morning or afternoon, for 12 weeks, found the largest effect sizes for the five-times sit-to-stand and for drowsiness in the afternoon NMN group (Kim 2022). Every group in that trial, placebo included, improved on sit-to-stand, so the best hour is not settled.
Give it weeks. Blood NAD+ rose early: in an 8-week trial in healthy overweight adults, 100, 300 and 1,000 mg of NR a day raised whole-blood NAD+ by 22%, 51% and 142% within two weeks, and the rise held for the rest of the study (Conze 2019). In a 14-day randomized trial in 65 healthy adults, NMN and NR each raised circulatory NAD+ (Christen 2026). The trials that measured walking, sleep or biological age ran about 8 to 12 weeks, so that is the window the evidence covers.
What the trials measured beyond NAD+. In a 60-day trial of 80 middle-aged adults, six-minute walking distance rose more on 300 to 900 mg of NMN than on placebo, self-rated health on the SF-36 improved, and blood biological age held steady while it rose in the placebo group (Yi 2023). In older adults on 250 mg a day, 4-metre walking time was shorter and sleep quality better than on placebo at 12 weeks, though the main stepping test did not differ (Morifuji 2024). A 2026 meta-analysis found no significant effect on body weight, fasting glucose, lipids or systolic blood pressure (Yang 2026). Across all the human trials, a 2026 systematic review found results on function and other healthspan outcomes heterogeneous and often null (Gallagher 2026).

Who the trials didn’t cover
The trials enrolled adults. Pregnant or breastfeeding women and people under 18 were not studied. If you take prescription medicine, check with your clinician first.
What’s in the product
NMN Liposomal Capsules give 500 mg of NMN from two capsules over 45 servings. Renue says the liposomes “protect the active ingredient through digestion for a slower, more sustained release,” and the 500 mg serving sits inside the 250 to 2,000 mg a day the human trials used.
NMN Pure Powder is 500 mg per scoop with 200 servings and no other ingredients. Renue suggests half a scoop twice daily for the first week if you are new to NMN, which is the start-small step a beginner is looking for.
NR Powder Smooth Taste Blend is a 1,000 mg serving of nicotinamide riboside, the top dose in the Conze 2019 trial. Renue says “NR is one of the most studied ways to support healthy NAD+ levels.”
For several precursors in one capsule, NAD+ Complete is a 4-in-1 liposomal blend of NMN, NR, trigonelline and NAD+ for slower release.

Questions readers ask
- What dose of NMN do the human trials use?
- Most used 250 to 900 mg a day: 250 mg a day in three trials and 300, 600 and 900 mg in another, where blood NAD+ and walking distance peaked at 600 mg. A 2026 meta-analysis covered doses from 250 to 2,000 mg a day.
- Does NMN actually raise NAD+ in people?
- Yes. In a 14-day randomized trial in 65 healthy adults, NMN and NR both raised circulatory NAD+, while nicotinamide did not. Source PubMed 41540253
- When should I take NMN?
- In the one trial that compared timing, the afternoon NMN group showed the largest sit-to-stand and drowsiness effects in older adults. Every group improved on sit-to-stand, so the best hour is not settled. Source PubMed 35215405
- Will I feel a difference from NMN?
- Some trials found one, such as longer or faster walks and better sleep quality. Across all human trials, a 2026 systematic review found function results heterogeneous and often null.
- Is NMN safe to take every day?
- Within the durations tested: 1,250 mg a day for up to four weeks caused no severe adverse events in a placebo-controlled trial of 31 healthy adults. Source PubMed 36002548
